Retatrutide cleared its first late-stage study at 23.7% weight loss

Adults with obesity and knee osteoarthritis on the highest dose of retatrutide lost an average of 23.7% of their body weight at 68 weeks counting everyone who enrolled, and 28.7% among those who stayed on it. Knee pain fell by up to 62.6% on average, and more than 1 in 8 participants finished with no knee pain at all.

Retatrutide mimics three hunger-regulating hormones, GLP-1, GIP and glucagon, where the injections already in use work on one or two, and higher doses of tirzepatide have reduced average body weight by roughly 20.9% in late-stage studies. TRIUMPH-4 was not built around weight alone, so trials designed for that could read differently, and it is still unclear when the drug could reach the market. Seven more phase three trials are expected to report by the end of 2026.

Tolerability is where the picture gets more complicated. Roughly 18% of participants on the highest dose stopped treatment early against 4% on placebo, and the company tied those exits to starting body mass index and to what it called perceived excessive weight loss; among people who began at a BMI of 35 or higher, 12% stopped. Which dose, and whether any of it suits a particular body, belongs with a qualified clinician.
